The band’s performance at the event, which runs 17-20 July 2014 at Henham Park, is expected to be their only UK gig of the year.
Billy Bragg, Anna Calvi, Röyksopp and Californian pop punk act Haim are also among the first wave of artists to be named on the bill.
Kevin Baird from Two Door Cinema Club said: ‘It's really exciting for us to be headlining Latitude in 2014. Over the past five years we've made the progression from playing in the small tents to being close to the top of the bill on the main stage.
‘Headlining a great festival like Latitude is obviously quite a big deal for us but we also feel like it is the next natural step. We're ready to headline. We're ready to smash it.’
